Hyun In-taek's statement comes a day after the DPRK re-opened the border on Tuesday. Pyongyang had closed it on Friday in protest at the joint South Korea-US military exercises.
South Korea has warned that it will respond with decisive action if Pyongyang again blocks access to the complex.
The Unification Ministry says over a hundred South Korean firms were operating in the Kaesong complex by the end of January.
