Johnson will be in charge of an 11 billion pound budget covering public transport, police and fire services in a city of some 7.5 million people. He will also oversee preparations for the London 2012 Olympic Games.
The outcome followed a catastrophic result for Prime Minister Brown and his governing Labor Party in local elections across England and Wales. Brown was forced to make a pledge to heed the harsh verdict from voters, after his party lost London's City Hall and more than 300 municipal council seats.
